چکیده

The penamaldic derivatives of amoxicillin, ampicillin, and penicillins G and V, stabilized with Zn(2+), were obtained from a methanolic medium. The enzymatic kinetic results show that the these derivatives elicit reversible inhibition of the enzyme metallo-beta-lactamase from Bacillus cereus, with inhibition constant values determined at pH 7.0 and 25 degrees C.

Inhibition of metallo-beta-lactamases by a series of mercaptoacetic acid thiol ester derivatives.

A series of mercaptoacetic acid thiol esters have been identified as metallo-beta-lactamase inhibitors. Electrospray mass spectrometry (ESMS) has shown that irreversible inhibition of the Bacillus cereus II metallo-beta-lactamase by SB214751, SB214752, and SB213079 was concomitant with a 90-Da increase in mass of the enzyme. Tryptic digestion of the B. cereus II inhibited with SB214751 illustra...

Characterization and sequence of the Chryseobacterium (Flavobacterium) meningosepticum carbapenemase: a new molecular class B beta-lactamase showing a broad substrate profile.

The metallo-beta-lactamase produced by Chryseobacterium (formerly Flavobacterium) meningosepticum, which is the flavobacterial species of greatest clinical relevance, was purified and characterized. The enzyme, named BlaB, contains a polypeptide with an apparent Mr of 26000, and has a pI of 8.5. It hydrolyses penicillins, cephalosporins (including cefoxitin), carbapenems and 6-beta-iodopenicill...

Processing of Bacillus cereus 569/H beta-lactamase I in Escherichia coli and Bacillus subtilis.

The gene for Bacillus cereus 569/H beta-lactamase I, penPC, has recently been cloned and sequenced (Mézes, P. S. F., Yang, Y. Q., Hussain, M., and Lampen, J. O. (1983) FEBS Lett. 161, 195-200). A typical prokaryotic signal peptide but with no lipoprotein modification site, as present in the Bacillus licheniformis 749/C beta-lactamase, was indicated by the DNA sequence for this secretory protein...

Thiol-group binding of zinc to a beta-lactamase of Bacillus cereus: differential effects on enzyme activity with penicillin and cephalosporins as substrates.

Zinc, which is required for the hydrolysis of cephalosporins by a crude enzyme from Bacillus cereus 569, also increased the stability of this activity during storage. A loss in activity of the zinc-activated enzyme which occurred on prolonged hydrolysis of cephalosporin C was not restored by further addition of zinc. The thiol reagents N-ethyl maleimide (NEM), iodoacetic acid (IAA), CdCl(2), an...

Comparison of Activity in Culture Medium

The aminoglycoside antibiotics, particularly gentamicin, are increasingly widely used and frequently in patients with impaired renal function. All these drugs are potentially ototoxic and measurement of blood levels is often desirable but may be complicated by the presence of a second drug, most commonly a penicillin or cephalosporin. Assays of streptomycin in the presence of penicillin have be...
